My (new to me) 2000 400DB has a E140 multifunction display and a RayPilot 650. Should they talk to each other for route following or are they incompatible? I think they should be SeaTalk compatible but the E140 gave an error message that there was no Seatalk connection. Trying to figure out where to start diagnosing this.

Is the E140 and raypilot on the seatalk network? The raypilot has 2 connections on the back for seatalk1. Hopefully one goes straight to the E140. The other one should go to the course computer. This is not mandatory though.

I think the issue is with the E140 as I get a Seatalk error message on startup.

I would swap the seatalk cable with a known good one. Might be that easy.
